建立了测定熊胆中3种主要磷脂组分即磷脂酰胆碱(PC)、磷脂酰肌醇(PI)、磷脂酰乙醇胺(PE)的高效液相色谱法。将熊胆风干后研成粉末,经V(氯仿)vV(甲醇)= 1v1提取液处理后,在P-ESilica柱上进行HPLC分析。流动相为V(乙腈)vV(甲醇)= 76v24,流速1.5 m L/m in,紫外检测波长205 nm 。PC的平均回收率为89.30% ,相对标准差RSD = 2.0% 。在精密度实验中,PI,PC,PE保留时间的RSD 分别为3.9% ,1.2% ,1.9% ,峰面积的RSD分别为1.6% ,0.89% ,2.6% 。所建立的方法可作为熊胆中磷脂类化合物PC,PI,PE的一种简便、快速、准确的测定方法。
A high performance liquid chromatography (HPLC) was developed for the determination of phosphatidylcholine (PC), phosphatidylinositol (PI) and phosphatidylethanolamine (PE) The bear gall was air-dried and ground into powder. After V (chloroform) vV (methanol) = 1 v1 extract, HPLC analysis was performed on a P-ESilica column. The mobile phase was V (acetonitrile) vV (methanol) = 76v24, the flow rate was 1.5 m L / min and the UV detection wavelength was 205 nm. The average recovery rate of PC was 89.30%, relative standard deviation RSD = 2.0%. In the precision experiments, the RSDs of retention time of PI, PC and PE were 3.9%, 1.2% and 1.9%, respectively. The RSDs of peak areas were 1.6% and 0.89%, respectively. 6%. The established method can be used as a simple, rapid and accurate method for the determination of phospholipids PC, PI and PE in bile.
