Objective Leptin, a polypeptide secreted and synthesized by adipocytes, negatively reduces appetite and enhances energy metabolism to maintain body fat balance. In the fetus and neonatal period, placenta and mammary gland synthesis and secretion of leptin, fetal and neonatal development and maturation play a regulatory role. This study explored the detection of breast-milk leptin and explored the dynamic changes of the content of breast-milk leptin and its impact on the level of leptin in neonates. Methods Whole milk was collected 5-10 ml (1 week postpartum breast milk) and mixed to prepare whole milk. The whole milk was centrifuged to remove skim milk to make skim milk. The whole milk and skim milk were centrifuged by ultrasonic vibration and the liquid phase and radiation Immune detection of leptin content. Neonatal femoral vein blood, serum, radioimmunoassay leptin content. Results The leptin content in whole milk was (53.9 ± 29.1) ng / ml in 60 cases of breast milk, (6.2 ± 8.3) ng / ml in whole milk after defatted, and the content of whole milk and defatted milk leptin was significantly Difference (t = 8.28, P <0.01). Peripheral blood leptin level was (2.8 ± 1.2) ng / ml in 42 cases of exclusively breastfeeding neonates, and peripheral blood leptin level was (1.9 ± 0.9) ng / ml in 36 cases of pure formula-fed neonates. There was significant difference between the two groups Significant difference (t = 3.37, P <0.01). Conclusion Breast milk contains leptin and is mainly associated with fat globules. Breastfeeding can increase leptin level in neonatal circulatory blood flow and play a positive role in neonatal adaptation to extrauterine environment.
