目的:检测nm 23 H1 与m uc1 基因在乳腺癌中的表达水平,探讨nm 23 H1 与m uc1 在乳腺癌原发灶与转移灶中表达水平改变的临床意义. 方法:采用定量 R T P C R 与图像扫描定量分析技术检测22 例家族性乳腺癌及60 例散发性乳腺癌原发灶与转移灶中nm 23 H1 与 m uc1 基因的表达水平.结果:①在22 例有家族史的原发灶中, nm 23 H1 基因在10 例无转移的标本中检出率为8/10,12 例转移淋巴结中检出率4/12,癌旁组织中检出率为14/22;60 例散发性乳腺癌中,nm 23 H1 基因在40 例无转移的原发灶中检出率为 34/40,20 例转移淋巴结中检出率为7/20,癌旁组织中检出率为39/60;无转移的原发灶与转移灶中的检出率存在显著性差异( P< 001);②nm 23 H1 基因在乳腺癌原发灶呈高表达,在转移灶中呈低表达,表达量在两者之间存在显著性差别( P< 0.001);nm 23 H1 基因表达水平的高低与乳癌的分级、病理类型、家族史无关;③ m uc1 在22 例有家族史的无转移原发灶中检出率为2/10,转移淋巴结中检出率为10/12,癌旁组织中检出率为 15/22;60 例散发
Objective: To detect the expression levels of nm23H1 and muc1 genes in breast cancer, and to explore the clinical significance of the changes of nm23H1 and muuc1 expression levels in primary and metastatic breast cancers. Methods: The quantitative expression of nm23H1 and muc1 genes in primary and metastatic breast cancers of 22 familial breast cancers and 60 sporadic breast cancers were detected by quantitative R T P C R and image scanning quantitative analysis techniques. Results: 1 Among the 22 primary lesions with family history, the detection rate of nm23H1 gene was 8/10 in 10 specimens without metastasis, and the detection rate was 4/12 in 12 metastatic lymph nodes. The detection rate in tissue was 14/22; among the 60 sporadic breast cancers, the detection rate of nm23H1 gene was 40/40 in 40 primary metastases without metastases, and the detection rate was 20 in metastatic lymph nodes. 7/20, the detection rate in adjacent tissues was 39/60; there was a significant difference in the detection rate between primary tumors and metastases without metastasis (P<0.01); 2nm 23H1 gene in breast cancer The primary tumors were highly expressed and were lowly expressed in metastases.
