目的:建立一种血红蛋白氧载体中巯基抗氧化剂的方法。方法:研究发现巯基抗氧化剂和聚合血红蛋白之间有一定的吸附作用并且难以分离,因此以血红蛋白为对照品,邻二氮菲-Fe3+为显色剂和氧化剂,在510nm处直接测定分别含有半胱氨酸、谷胱甘肽和氮-乙酰-半胱氨酸的3种血红蛋白氧载体溶液中这3种巯基抗氧化剂的含量。结果:该方法在1×10-6~1.5×10-5mol.L-1范围内有良好的线性关系,相关系数分别为0.9991,0.9989,0.9985,RSD分别为4.1%,3.4%,3.9%,回收率分别为97.3%±3.5%,96.7%±4.1%,95.5%±4.3%。结论:该方法避免了聚合血红蛋白与巯基抗氧化剂的分离并可直接测定血红蛋白氧载体中巯基抗氧化剂含量,是一种简便、准确、回收率高的方法。

Conclusion: This method can avoid the separation of polymeric hemoglobin and sulfhydryl antioxidants and directly determine the content of sulfhydryl antioxidants in hemoglobin oxygen carrier. This method is a simple, accurate and high recovery method.
