目的调查2011年襄阳市手足口病主要病原EV71的基因型特征,分析和探讨该型病毒的VP1基因变异和分子进化特点。方法对2011年襄阳市流行株EV71进行VP1区核苷酸序列测定和同源性比较及遗传进化分析。结果 2011年襄阳市手足口病EV71病毒的VPl区核苷酸序列全长均为891bp,与对照EV71毒株核苷酸序列同源性为96.5%~99.1%,编码蛋白氨基酸序列同源性为98.1%~100%。VP l区基因遗传进化分析显示,该中EV71病毒属于C4a基因亚型。结论 2011年襄阳市手足口病疫情主要病原EV71病毒均属于C4a基因亚型,未产生明显的抗原漂移及变异。
Objective To investigate the genotypes of EV71, the major pathogen of hand, foot and mouth disease in Xiangyang in 2011, and to analyze and explore the VP1 gene mutation and molecular evolution characteristics of this virus. Methods The VP1 region of EV71 in Xiangyang City in 2011 was sequenced and its homology and genetic evolution were analyzed. Results The full-length nucleotide sequence of VP1 of EV71 virus in Xiangyang City was 891bp in 2011, which shared 96.5% -99.1% identity with that of control EV71 strain. The homology of amino acid sequence of the encoded protein was 98.1% ~ 100%. Genetic analysis of the VP1 gene revealed that the EV71 virus belongs to the C4a genotype. Conclusion The major EV71 viruses of hand, foot and mouth disease in Xiangyang City in 2011 belonged to subtype C4a, which did not produce obvious antigen drift and mutation.
