目的建立一种从健康人脐带中分离间充质干细胞(mesenchymal stem cells,MSCs)的简单、经济、高效方法,并探讨其对小鼠免疫细胞的影响。方法取经剖宫产获得的健康人脐带血管与外膜之间的胶状物并剪切成小于1 mm3的小块,用组织块贴壁法直接培养MSCs,并用获得的MSCs注射于健康C57 BL/6小鼠皮下。结果获得的MSCs高表达CD105、CD73、CD44和CD90,不表达CD34、CD45和人类白细胞抗原-DR,符合干细胞的各种生物学特性。结论用简便、经济的方法获得了健康人脐带间充质干细胞,注射C57 BL/6小鼠体内后,小鼠的血细胞及T细胞亚型未改变,自身免疫系统稳定,为进一步的研究奠定了实验基础。
Objective To establish a simple, economical and efficient method for the isolation of mesenchymal stem cells (MSCs) from umbilical cord of healthy individuals and to investigate its effect on immune cells in mice. Methods The cords between the umbilical cord and adventitia were obtained from cesarean section and cut into small pieces less than 1 mm3. MSCs were cultured directly by tissue block method and injected into healthy C57 BL / 6 mice subcutaneously. Results The MSCs were highly expressed CD105, CD73, CD44 and CD90, but not CD34, CD45 and human leukocyte antigen-DR, which accorded with various biological characteristics of stem cells. Conclusion The human umbilical cord mesenchymal stem cells were obtained in a simple and economical way. After being injected into C57 BL / 6 mice, the blood cells and T cell subtypes of mice did not change and the autoimmune system was stable, which laid the foundation for further research Experimental basis.
