目的研究抗重组日本血吸虫生殖相关蛋白WD40(SjWD40)多克隆抗体对天然SjWD40的结合活性,观察SjWD40在虫体内的定位。方法从阳性钉螺体内逸出尾蚴,感染家兔,于感染后42 d剖杀收集成虫和虫卵制备石蜡切片,利用抗重组蛋白SjWD40多克隆抗体,通过间接免疫荧光方法探讨SjWD40在虫体内的分布。结果SjWD40广泛分布于雌虫的卵黄腺、雄虫的睾丸,虫卵的毛蚴体壁,尾蚴的头腺、钻腺、腺管等部位。结论免疫荧光染色法确定SjWD40蛋白主要分布于血吸虫成虫的生殖腺、毛蚴体壁及尾蚴腺体等组织。
Objective To study the binding activity of SjWD40 to the natural SjWD40 against the recombinant SjWD40 polyclonal antibody of recombinant Schistosoma japonicum and to observe the localization of SjWD40 in the parasite. Methods The cercariae were infected from the positive snails and infected by rabbits. The collected parasite and paraffin sections were collected at 42 days after infection to prepare paraffin sections. The anti-SjWD40 polyclonal antibody was used to investigate the distribution of SjWD40 in the parasite by indirect immunofluorescence . Results SjWD40 is widely distributed in the female yolk gland, male testis, egg larvae, cercariae of the head gland, gland, duct and other parts. Conclusion Immunofluorescence staining confirmed that the SjWD40 protein mainly distributed in the gonads of the schistosomiasis gonads, the body wall of the larvae and cercariae.
