JWA基因是受维甲酸(RA)、13顺-维甲酸(13-cRA)和佛波酯(TPA)等调控的细胞骨架相关基因.以前的研究发现,JWA基因与细胞分化和凋亡有关.为了探讨JWA基因在急性早幼粒细胞性白血病(APL)细胞分化和凋亡过程中的作用及机制,分别用ATRA,4HPR,TPA和As2O3处理NB4细胞,Western blot和RT-RCR检测JWA基因在蛋白和mRNA水平的表达,同时检测细胞周期和CD11b,CD33细胞表面抗原,分析细胞分化和凋亡.结果表明ATRA和As2O3分别诱导细胞分化和凋亡,而4HPR和TPA对NB4细胞分化和凋亡有双重诱导作用.在诱导细胞分化和凋亡过程中,JWA基因的表达水平均上调,但PML/RARα融合基因变化不明显;用JWA反义核酸处理NB4细胞后,TPA诱导其分化和调亡的作用受到明显抑制.TPA对NB4的作用可能是通过由JWA参与的直接和间接两种途径实现的.在对APL(M3)原代细胞的研究中除观察到与NB4细胞相似的JWA的变化外,还发现一特异的与APL细胞分化和凋亡有关的异常分子量的JWA蛋白;异常分子量的JWA蛋白是否是APL(M3)区别于NB4细胞的一种分子标志物以及JWA是否通过caspases信号调节通路参与NB4细胞分化和凋亡等均有待进一步研究证实.
The JWA gene is a cytoskeleton-related gene regulated by retinoic acid (RA), 13-cis-retinoic acid (13-cRA) and phorbol ester (TPA). Previous studies have found that JWA gene is involved in cell differentiation and apoptosis. To investigate the role and mechanism of JWA gene in the differentiation and apoptosis of acute promyelocytic leukemia (APL) cells, NB4 cells were treated with ATRA, 4HPR, TPA and As2O3, respectively. Western blot and RT-RCR were used to detect the JWA gene. Protein and mRNA levels were measured simultaneously with cell cycle and CD11b, CD33 cell surface antigens analyzed for cell differentiation and apoptosis. Results showed that ATRA and As2O3 induced cell differentiation and apoptosis, respectively, whereas 4HPR and TPA induced NB4 cell differentiation and apoptosis. There is a dual induction effect. In the process of inducing cell differentiation and apoptosis, the expression of JWA gene is up-regulated, but the change of PML/RARα fusion gene is not obvious; after treatment of NB4 antisense nucleic acid, TPA induces its differentiation and apoptosis. The effect of TPA was significantly inhibited. The effect of TPA on NB4 may be achieved through the direct and indirect routes involved by JWA. In addition to the observation of changes in JWA similar to NB4 cells in the study of APL (M3) primary cells.
