目的:探讨NDRG1在胰腺癌细胞中的表达及其与MMP-7的关系。方法:检测NDRG1与MMP-7蛋白与m RNA在4种胰腺癌细胞株(PANC-1、BXPC-3、CAPAN-2、SW1990)表达;构建靶向NDRG1的干扰质粒si NDRG1,转染PANC-1细胞,检测转染后PANC-1细胞NDRG1与MMP-7蛋白与m RNA的表达,以及增殖与调亡情况。结果:4种细胞株中均有NDRG1与MMP-7蛋白与m RNA的表达,且两者的表达水平随着细胞分化程度的降低而升高(均P<0.05);PANC-1细胞转染si NDRG1后,NDRG1与MMP-7的蛋白与m RNA表达均明显降低、细胞增殖明显抑制、凋亡率明显升高(均P<0.05)。结论:NDRG1的表达与胰腺癌细胞的分化程度密切相关,且可能通过上调MMP-7表达促进胰腺癌细胞的生长。
Objective: To investigate the expression of NDRG1 in pancreatic cancer cells and its relationship with MMP-7. Methods: The expression of NDRG1 and MMP-7 protein and m RNA were detected in four pancreatic cancer cell lines (PANC-1, BXPC-3, CAPAN-2 and SW1990). The NDRG1 targeting NDRG1 plasmid was constructed and transfected into PANC- 1 cells. The expression of NDRG1, MMP-7 protein and m RNA in PANC-1 cells and the proliferation and apoptosis were detected. Results: The expression of NDRG1, MMP-7 protein and m RNA in both cell lines were significantly higher than those in control group (P <0.05). PANC-1 cells were transfected si NDRG1, NDRG1 and MMP-7 protein and m RNA expression were significantly reduced, cell proliferation was significantly inhibited, the apoptosis rate was significantly increased (all P <0.05). Conclusion: The expression of NDRG1 is closely related to the differentiation of pancreatic cancer cells and may promote the growth of pancreatic cancer cells by up-regulating the expression of MMP-7.
