目的:研究不同周龄ApoE~(-/-)雄性小鼠离体胸主动脉内皮依赖性舒张功能的变化。方法:采用不同周龄(18~44周)ApoE~(-/-)雄性小鼠,分离其胸主动脉,以不同浓度的乙酰胆碱(Ach)分别评价其内皮的完整性,并绘制浓度-舒张曲线,观察不同周龄ApoE~(-/-)小鼠胸主动脉内皮的依赖性舒张作用;胸主动脉行油红O染色,以评价不同周龄ApoE~(-/-)小鼠胸主动脉脂质沉积、斑块面积大小。结果:不同周龄ApoE~(-/-)小鼠离体胸主动脉在浓度为10-5mol/L的Ach作用下,其舒张率均<80%,且随着周龄的增加,舒张率呈逐渐降低趋势;与24周相比,48周ApoE~(-/-)雄性小鼠离体胸主动脉在不同浓度Ach下,其舒张率均显著降低;与24周和36周ApoE~(-/-)小鼠相比,48周ApoE~(-/-)小鼠其斑块面积/血管剖面面积显著增加。不同周龄ApoE~(-/-)小鼠血清中总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)差异均无统计学意义(P>0.05)。ApoE~(-/-)雄性小鼠胸主动脉内皮完整性、血管舒张功能与周龄呈负相关关系(P<0.05),同时与脂质沉积、斑块面积也呈负相关关系(P<0.05)。结论:ApoE~(-/-)雄性小鼠血管内皮依赖性舒张率随着周龄及斑块面积/血管剖面面积的增加而降低,其胸主动脉血管内皮依赖性舒张功能与周龄、斑块面积/血管剖面面积呈负相关关系。
AIM: To study the changes of endothelium-dependent vasodilatation in isolated thoracic aorta of ApoE ~ (- / -) male mice of different weeks of age. Methods: Thoracic aorta was isolated from male ApoE ~ (-) mice of different weeks of age (18 ~ 44 weeks). The integrity of endothelium was evaluated by different concentrations of acetylcholine (Ach) Curve to observe the endothelium-dependent relaxation of thoracic aorta of ApoE ~ (- / -) mice at different weeks of age. The aorta of the thoracic aorta was stained with oil red O to evaluate the expression of thoracic apoE ~ (- Arterial lipid deposition, plaque area size. Results: In vitro ApoE ~ (- / -) mouse thoracic aorta in the concentration of 10-5mol / L of Ach, the diastolic rate was <80%, and with increasing age, diastolic rate (P <0.05). Compared with 24 weeks, the relaxation rate of isolated thoracic aorta of ApoE ~ (- / -) male mice in 48 weeks was significantly decreased at different concentrations of Ach; Compared with 24 weeks and 36 weeks of ApoE ~ Week-old ApoE ~ (- / -) mice showed a significant increase in plaque area / vascular cross-sectional area. There was no significant difference in serum TC, TG, HDL, LDL between ApoE ~ (- / -) mice at different weeks of age (P > 0.05). The aortic endothelium integrity and vasodilatation in ApoE ~ (- / -) male mice were negatively correlated with age (P <0.05), and negatively correlated with lipid deposition and plaque area (P < 0.05). CONCLUSION: The endothelium-dependent relaxation rate of ApoE ~ (- / -) male mice decreases with the increasing of age and plaque area / vascular cross-sectional area. The endothelium-dependent vasodilatation of thoracic aorta is related to age, Block area / vascular cross-sectional area was negatively correlated.
