目的:研究五味子醇甲对β-淀粉样蛋白(amyloidβ-protein,Aβ)损伤大鼠肾上腺髓质嗜铬瘤细胞(PC12细胞)的预保护及治疗作用。方法:体外培养PC12细胞,建立Aβ引起的细胞损伤模型;采用细胞形态学观察突起长度及神经网络的变化,细胞形态学定量测定有突起的细胞数目,MTT法测定细胞活性。结果:细胞形态学、细胞形态学定量和细胞活性测定结果表明,与Aβ模型组比较,五味子醇甲低、中、高剂量(1、3、9μg/mL)组PC12细胞神经突起较长,神经网络破坏的程度较轻,有突起细胞数目增多,细胞活性增高。结论:五味子醇甲具有拮抗Aβ神经毒性的作用,对Aβ损伤PC12细胞具有预保护及治疗作用。
Objective: To study the protective and therapeutic effects of schizandrin on the adrenal medullary pheochromocytoma (PC12) cells in amyloid β-protein (Aβ) -induced injury in rats. Methods: PC12 cells were cultured in vitro to establish Aβ-induced cell injury model. Cell morphology was used to observe the length and neural network changes. Cell morphology was used to quantitatively determine the number of protruding cells. Cell viability was measured by MTT assay. Results: The results of cell morphology, cytomorphometry and cell viability assay showed that compared with Aβ model group, PC12 cells in low, medium and high doses of 1, 3 and 9μg / mL of Schisandra alcohol group had longer neurites, The extent of network damage is lighter, with an increase in the number of protruding cells, increased cell viability. CONCLUSION: Schisandrae A has the neuroprotective effect against Aβ and has the protective and therapeutic effects on Aβ-injured PC12 cells.
