应用流式细胞术和细胞免疫荧光染色技术,对甲状腺腺瘤和甲状腺癌细胞p16基因蛋白表达进行定量研究。结果表明,甲状腺癌p16蛋白表达量明显低于甲状腺腺瘤(P<001);p16蛋白表达随甲状腺恶性程度的增高而下降(P<005)。提示p16基因失活与甲状腺癌发生有关,p16蛋白表达缺失与肿瘤分化不良有关,可作为临床判断肿瘤分化程度和预后的参考指标之一。
The expression of p16 gene protein in thyroid adenoma and thyroid cancer cells was quantitatively studied by flow cytometry and immunofluorescence staining. The results showed that the expression of p16 protein in thyroid carcinoma was significantly lower than that in thyroid adenoma (P<001); the expression of p16 protein decreased with the increase of thyroid malignancy (P<005). It is suggested that the inactivation of p16 gene is related to the occurrence of thyroid cancer. The loss of p16 protein expression is associated with poor tumor differentiation. It may be used as one of the reference indexes for clinical judgment of tumor differentiation and prognosis.
