目的:考察河北不同产地知母药材中芒果苷及菝葜皂苷元的含量。方法:分别采用HPLC法和HPLC-ELSD法测定芒果苷和菝葜皂苷元的含量。测定芒果苷以水∶乙腈∶0.85%磷酸(100∶25∶5)为流动相;流速为0.6mL/min;柱温30℃;检测波长为258nm。测定菝葜皂苷元以甲醇∶水(95∶5)为流动相;流速为1.0mL/min;柱温30℃;用蒸发光散色检测器检测,漂移管温度85℃。结果:河北不同产地知母药材中菝葜皂苷元含量为1.51%~1.73%,芒果苷含量为0.11%~0.73%。结论:本法测定知母药材中菝葜皂苷元及芒果苷含量,方法简单、快速、准确、重现性好,方法学考察符合含量测定的要求,可以用来控制知母药材质量。
Objective: To investigate the content of mangiferin and saponin in Astragalus membranaceus from different producing areas in Hebei Province. Methods: The contents of mangiferin and saponin were determined by HPLC and HPLC-ELSD respectively. The mangiferin was assayed with water:acetonitrile:0.85% phosphoric acid (100:25:5) as mobile phase; flow rate was 0.6 mL/min; column temperature was 30°C; detection wavelength was 258 nm. Determination of sarsasapogenin methanol: water (95:5) as the mobile phase; flow rate of 1.0mL/min; column temperature 30 °C; detected by evaporative light scattering color detector, drift tube temperature 85 °C. Results: The content of sarsasapogenin in Radix Astragali from different producing areas in Hebei Province was 1.51%-1.73%, and the content of mangiferin was 0.11%-0.73%. Conclusion: This method determines the content of saponins and mangiferin in Zhimu herbs. The method is simple, rapid, accurate and reproducible. The methodological study meets the requirements of content determination and can be used to control the quality of Zhimu medicinal materials.
