目的观察麝香保心丸对急性心肌梗死模型大鼠血流动力学及血浆炎性因子的影响,探讨其改善急性心肌梗死后心室重构的机制。方法 SD大鼠70只,选取8只为空白对照组,其余采用结扎冠状动脉左前降支的方法造成急性心肌梗死模型,造模成功大鼠随机分为假手术组8只、模型组、麝香保心丸等剂量组、麝香保心丸大剂量组及辛伐他汀组各9只。空白对照组、假手术组和模型组给予生理盐水,辛伐他汀组给予辛伐他汀片每天0.2mg/100g,麝香保心丸等剂量、大剂量组分别给予麝香保心丸每天1.4mg/100g、2.4mg/100g,各组大鼠均灌胃给药。2周后,测定各组大鼠血流动力学指标,并检测血浆白细胞介素6(IL-6)和肿瘤坏死因子-α(TNF-α)的含量。结果与空白对照组相比,模型组的左室收缩末压(LVESP)、左室内压最大上升速率(LV+dp/dtmax)和左室内压最大下降速率(LV-dp/dtmax)显著降低(P<0.01),左室舒张末压(LVEDP)显著增加(P<0.01),血浆IL-6与TNF-α含量显著升高(P<0.01);与模型组比较,麝香保心丸等剂量、大剂量组的LVESP、LV+dp/dtmax、LV-dp/dtmax显著增加(P<0.05),LVEDP显著下降(P<0.01),血浆IL-6与TNF-α含量显著降低(P<0.05或P<0.01)。结论麝香保心丸能改善模型大鼠急性心肌梗死后血流动力学参数,并减少血浆中炎性因子的释放,这可能是其改善模型大鼠急性心肌梗死后心室重构的机制。
Objective To observe the effect of Shexiang Baoxin Pill on the hemodynamics and plasma inflammatory factor in acute myocardial infarction (AMI) rats and to explore the mechanism of its improving the ventricular remodeling after acute myocardial infarction. Methods Seventy SD rats were randomly divided into sham operation group (n = 8), control group (n = 8) and left anterior descending coronary artery (ligation) Heart pills and other dose groups, high-dose group of musk heart pill and simvastatin group of 9. The rats in the blank control group, the sham operation group and the model group were given physiological saline. Simvastatin tablets were administrated with 0.2 mg / 100 g simvastatin tablets every day. The other three doses of Shexiang Baoxin Pill were given 1.4 mg / 100 g , 2.4mg / 100g, each group rats were intragastric administration. After 2 weeks, the hemodynamics of the rats in each group were measured, and the levels of plasma interleukin 6 (IL-6) and tumor necrosis factor-α (TNF-α) were measured. Results Compared with the blank control group, LVESP, LV + dp / dtmax and LV-dp / dtmax of model group were significantly decreased (P <0.01), left ventricular end diastolic pressure (LVEDP) increased significantly (P <0.01), plasma IL-6 and TNF-αlevels significantly increased (P <0.01); Compared with the model group, LVEDP, LV + dp / dtmax and LV-dp / dtmax increased significantly (P <0.05), LVEDP decreased significantly (P <0.01) and plasma IL-6 and TNF- Or P <0.01). Conclusion Shexiang Baoxin Pill can ameliorate the hemodynamic parameters of acute myocardial infarction rats and decrease the release of inflammatory cytokines in the model rats, which may be the mechanism of the improvement of ventricular remodeling after acute myocardial infarction.
