为探讨亚砷酸钠诱导NB_4细胞凋亡的作用机制,采用流式细胞术、DNA电泳和免疫印迹研究亚砷酸钠对NB_4细胞的作用。结果表明:①亚砷酸钠作用的NB_4细胞先阻滞在G_2+M期,而后发生凋亡;④dUTP-FITC特异性标记G_2+M期NB_4细胞,而且发生在G_2+M期阻滞后;③亚砷酸钠处理NB_4细胞的周期蛋白A、E、D_1、D_2和D_3无明显变化,周期蛋白B_1表达随作用时间的延长而增加;④流式细胞仪分析显示在亚砷酸钠处理的早期,有部分S期细胞表达周期蛋白B_1,晚期大部分高表达周期蛋白B_1细胞是处于G_2+M期,有少量亚倍体细胞高表达周期蛋白B_1。上述结果提示,亚砷酸钠通过上调周期蛋白B_1选择性地诱导G_2+M期NB_4细胞的凋亡。
To investigate the mechanism of apoptosis of NB_4 cells induced by sodium arsenite, flow cytometry, DNA electrophoresis and Western blotting were used to study the effect of sodium arsenite on NB_4 cells. The results showed that: 1 The NB_4 cells treated with sodium arsenite first arrested in the G_2+M phase, followed by apoptosis; 4dUTP-FITC specifically labeled G_2+M phase NB_4 cells, and occurred in the G_2+M phase arrest. 3 There were no significant changes in cyclin A, E, D_1, D_2, and D_3 in NB_4 cells treated with sodium arsenite, and the expression of cyclin B_1 increased with the prolonged action time; 4 Flow cytometry analysis showed that the treatment with sodium arsenite In the early stage, some S phase cells express cyclin B_1, and most of the high expression cyclin B_1 cells in the late phase are in the G_2+M phase, and a small number of subploid cells express high levels of cyclin B_1. The above results suggest that sodium arsenite selectively induces apoptosis of NB_4 cells in G_2+M phase by up-regulating cyclin B_1.
