目的:观察开心解郁汤对抑郁模型大鼠脑内杏仁核去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)含量的影响。方法:将30只SD雄性大鼠随机分为正常组、模型组、给药组,每组10只;用慢性应激结合孤养的方式造模21 d。在造模同时,给药组ig开心解郁汤40 g.kg-1,1次/d,连续21 d。用高效液相-电化学方法检测大鼠脑内NE,DA,5-HT含量的变化。结果:与正常组比较,模型组大鼠脑内NE,DA,5-HT含量明显减少;与模型组比较,开心解郁汤可显著升高抑郁模型大鼠脑内NE,DA,5-HT的含量(P<0.05)。结论:开心解郁汤的抗抑郁作用机制可能与调节脑内单胺类神经递质的平衡有关。
Objective: To observe the effect of Kaixin Jieyu Decoction on the contents of norepinephrine (NE), dopamine (DA) and 5-hydroxytryptamine (5-HT) in the amygdala of rat model of depression. Methods: Thirty male Sprague-Dawley rats were randomly divided into normal group, model group and administration group, with 10 rats in each group. The model rats were treated with chronic stress and solitary for 21 days. At the same time of modeling, the treatment group ig happy Jieyu Decoction 40 g.kg-1,1 times / d, for 21 days. The contents of NE, DA and 5-HT in rat brain were detected by HPLC-ECD method. Results: Compared with the normal group, the content of NE, DA and 5-HT in the model group decreased significantly. Compared with the model group, Kaixin Jieyu Decoction could significantly increase the levels of NE, DA and 5-HT (P <0.05). Conclusion: The antidepressant mechanism of Kaixin Jieyu Decoction may be related to the regulation of monoamine neurotransmitter in the brain.
