应用Northern杂交技术发现遗传听源惊厥易感大鼠P77PMC在出生14天后脑内c-fosmR-NA水平迅速增高,21天达到最高水平,而后逐渐下降,此与大鼠惊厥易感性形成规律相类似;惊厥后脑内c-fosmRNA具有快速、大量及一过性表达特点;惊厥前后15分钟内给予安定(10mg/kg)能有效阻断c-fos基因表达。推测c-fos基因间接地参与了P77PMC大鼠惊厥易感性的形成。
Northern blotting showed that the level of c-fosmR-NA in rat brain was significantly increased after 14 days of birth, reaching the highest level in 21 days and then gradually decreasing, which was similar to the formation of seizure susceptibility in rats The c-fos mRNA expression in brain was fast, large and transient after convulsion. Apo (10 mg / kg) within 15 minutes before and after convulsion could effectively block the expression of c-fos gene. It is speculated that c-fos gene indirectly involved in the formation of seizure susceptibility in P77PMC rats.
