目的在体记录海马CA1区场电位时,改良局部给药方式。方法动物麻醉后固定于立体定位仪上,参照立体定位参数将自制刺激/记录电极及给药套管分别插入海马,给药并记录CA1区局部场电位,而后进行LTP的诱发和记录。结果确定改良法给药部位的准确性及药物在海马内的扩散范围;用改良法局部给予溶剂对海马CA1区突触传递及LTP没有明显影响;用改良法局部给予侧脑室给药量1/10的Aβ1-42,即显著压抑了海马CA1区LTP,且不影响突触传递;用改良法局部给予溶剂或Aβ1-42对PPF没有明显影响。结论完善了海马CA1区在体刺激/记录及同侧海马局部给药的方法,且用药量小、操作简单;用改良法成功记录并观察了Aβ1-42对海马CA1区fEPSP、LTP及PPF的影响,实现了用该方法对神经突触可塑性的评价。
Objective To record the local field of CA1 in the hippocampus and improve the local administration mode. Methods The animals were fixed on the stereotactic instrument after anesthesia. The self-made stimulation / recording electrodes and the administration cannula were respectively inserted into the hippocampus with reference to the stereotaxic parameters. The local field potentials of the CA1 region were administered and recorded, and then the LTP induction and recording were performed. Results The accuracy of the improved method and the extent of drug diffusion in the hippocampus were determined. No significant effect of local administration of solvent on hippocampal CA1 synaptic transmission and LTP was observed. 10 of Aβ1-42, which significantly suppressed the hippocampal CA1 LTP, and does not affect the synaptic transmission; the local administration of the modified solvent or Aβ1-42 PPF no significant effect. Conclusion The method of local administration of ipsilateral hippocampal CA1 region in hippocampus is improved and the dosage is small and the operation is simple. The effect of Aβ1-42 on fEPSP, LTP and PPF Affect the realization of this method of synaptic plasticity evaluation.
