目的:通过观察糖脂平颗粒对糖尿病肾病大鼠肾组织一氧化氮(NO)、一氧化氮合酶(NOS)水平的影响。探讨糖脂平颗粒对糖尿病肾病大鼠肾脏的保护作用。方法:Wistar大鼠60只,雌雄各半,采用高脂饲料加小剂量(40 mg·kg-1)ip链脲佐菌素(STZ)造模,按体重编号,以随机数字表法分为4组:模型组、糖脉康颗粒组、糖脂平颗粒高、低剂量组,分别给与生理盐水、糖脉康颗粒、糖脂平颗粒进行干预,测定干预后各组大鼠肾组织匀浆NO和NOS,制备肾组织切片,观察肾组织形态学改变。结果:治疗后各治疗组NO为(37.24±4.16)~(44.74±6.08)μmol.g-1与模型组(30.47±8.33)μmol.g-1比较升高,NOS各治疗组为(0.44±0.13)~(0.61±0.17)U.mg-1与模型组(0.27±0.14)U.mg-1比较亦升高,均有显著性差异(P<0.05),糖脂平颗粒小剂量组与糖脉康颗粒组比较,指标均没有显著性差异;而糖脂平颗粒大剂量组与糖脉康颗粒组比较,NO及NOS均有显著性差异糖脂平颗粒高剂量组治疗效果优于低剂量组和糖脉康颗粒组。肾组织切片显示:糖脂平颗粒2个高、低剂量组和糖脉康颗粒组肾脏组织均有不同程度的修复,糖脂平颗粒高剂量组优于其他各给药组。结论:糖脂平颗粒可升高糖尿病肾病大鼠肾组织匀浆一氧化氮及NOS,同时促进肾脏受损组织的修复,对糖尿病肾脏具有保护作用
Objective: To observe the effects of Tangzhiping Granules on the levels of nitric oxide (NO) and nitric oxide synthase (NOS) in the kidney of diabetic nephropathy rats. To investigate the protective effect of Tangzhiping Granules on the kidney of diabetic nephropathy rats. Methods: Sixty male and female Wistar rats were randomly divided into three groups: high fat diet and low dose (40 mg · kg -1) of ip streptozotocin (STZ) The rats in model group, Tangmaikang granule group, and Tangzhiping granule high and low dose groups were given normal saline, Tangmaikang granule and Tangzhiping granule respectively. After intervention, the rats’ kidney homogenate NO and NOS were used to prepare kidney sections and observe the morphological changes of kidney. Results: Compared with the model group (30.47 ± 8.33) μmol.g-1, NO in the treatment group was significantly higher than that in the untreated group (37.24 ± 4.16 vs. 44.74 ± 6.08 μmol.g-1, (P <0.05). Compared with model group (0.27 ± 0.14) U.mg-1, U.mg-1 was significantly increased (P <0.05) Tangmaikang granule group, the indicators were no significant differences; and sugar-fat granule large dose group and the sugar veins Kang particles group, NO and NOS were significant differences in high-dose glycolipid treatment group was better than the low-dose Dose group and the Tangram group. Kidney tissue sections showed that the two high dose and low dose groups of Tangzhiping Granules and the Tongmaikang Granules group had different degrees of repair, and the high dose group of Tangzhiping Granules was superior to the other groups. CONCLUSION: Tangzhiping granule can increase nitric oxide and NOS in renal tissue of rats with diabetic nephropathy and promote the repair of damaged tissues in kidney, which has a protective effect on diabetic nephropathy
