目的 探讨化疗药物吉西他滨对人子宫颈癌 (HeLa)细胞系的放射增敏作用。方法 MTT法确定细胞抑制率≤ 1 0 %的吉西他滨 (dFdC)浓度 (IC1 0 ) ,用该浓度吉西他滨分别孵育细胞 4、8、1 2及 2 4h ,于弃药前、弃药后立刻及弃药后 1 2h ,分别行 2、4及 6Gy60 Coγ线照射。计数细胞存活分数 ,计算增敏比。结果 吉西他滨IC1 0 浓度为 0 .0 1 μmol/L ,该浓度吉西他滨分别作用于细胞 4、8、1 2及 2 4h ,弃药后立刻照射 ,增敏比分别为 1 .1 9、1 .35、1 .72、1 .93。作用 2 4h ,弃药前、弃药后立刻及弃药后 1 2h照射 ,增敏分别为 1 .91、1 .93、1 .5 2。结论 吉西他滨对HeLa细胞具有放射增敏作用。增敏作用随药物作用时间的延长而增加 ,持续作用短时间 (4h)就可产生放射增敏作用 ,作用 2 4h效果显著。弃药前照射及弃药后立刻照射 ,增敏效果好于弃药后 1 2h照射
Objective To investigate the radiosensitization effect of chemotherapeutic agent gemcitabine on human cervical carcinoma (HeLa) cell line. Methods MTT method was used to determine the concentration of gefitinib (dFcC) with cell inhibition rate ≤10% (IC10). The cells were incubated with gecitabine for 4, 8, 12 and 24 hours, respectively. Twelve and six hours after treatment, 2,4 and 6 Gy 60 Co gamma irradiation respectively. Count the cell viability score and calculate the sensitization ratio. Results Gemcitabine IC1 0 concentration of 0 1 μmol / L, the concentration of gemcitabine were applied to the cells 4,8,1 2 and 24 h, immediately after the discard of the drug, the sensitivities were 1.19, 1.35 , 1.72,1 .93. The role of 2 4h, abandoned before the drug, immediately after the abandonment of medicine and 1 2h after irradiation, sensitization were 1.91,1 .93,1 .5 2. Conclusion Gemcitabine has a radiosensitizing effect on HeLa cells. Sensitization increased with the extension of the role of drug duration, sustained effect of short-term (4h) can produce radiosensitization, the role of 24h significant effect. Abandoned drug irradiation and abandoned immediately after irradiation, sensitization better than after the drug 1 2h irradiation
