目的应用荧光原位杂交技术(FISH)直接检测慢性粒细胞白血病(CML)的融合基因(BCR/ABL)以辅助临床诊断和治疗白血病。方法应用FISH技术检测25例CML自1991年-2008年制备染色体剩余细胞悬液的BCR/ABL,6例是2008年的标本,7例是1991年-1995年的标本,余10例标本是2001年-2007年的。结果 25例CML患者经细胞遗传学检测费城染色体阳性Ph(+)23例,Ph(-)2例。FISH技术检测结果BCR/ABL阳性24例,BCR/ABL阴性1例。对照组5例为非CML的骨髓细胞,BCR/ABL均为阴性。结论 FISH技术能直接检测细胞中的融合基因(BCR/ABL),也可用于长期保存的细胞,其快速、简单、特异性高而备受青睐,是诊断和监测CML的一项有效的新技术,弥补了染色体检测报告需要时间长等的不足。
Objective To directly detect the fusion gene (BCR / ABL) of chronic myeloid leukemia (CML) by fluorescence in situ hybridization (FISH) to aid the clinical diagnosis and treatment of leukemia. Methods FISH was used to detect BCR / ABL in 25 cases of CML from 1991 to 2008, the remaining 8 cases of chromosomal suspension were prepared, 6 cases were in 2008, 7 cases were from 1991 to 1995, the remaining 10 cases were 2001 Year - 2007’s. Results Twenty - five patients with CML had Philadelphia Ph chromosome positive Ph (+) 23 cases and Ph (-) 2 cases by cytogenetics. FISH technology test results in 24 cases of BCR / ABL positive, BCR / ABL negative in 1 case. The control group of 5 cases of non-CML bone marrow cells, BCR / ABL were negative. Conclusion FISH can directly detect the fusion gene (BCR / ABL) in cells and can also be used for long-term preservation of cells. FISH is a fast, simple and highly specific method that is popular and is an effective new technique for diagnosis and monitoring of CML , Make up for the lack of chromosome test report takes a long time.
