目的:观察益气活血中药复方对CVB3病毒性心肌炎模型大鼠心肌细胞small inducible cytokine A2(MCP-1/CCL2)、ribosomal protein S4基因表达的影响,以期从基因水平揭示益气活血中药复方治疗CVB3心肌炎的作用机制,进一步证实益气活血中药复方是治疗CVB3病毒性心肌炎的有效方剂。方法:通过改良的抑制性消减杂交技术(SSH),克隆了受CVB3攻击的宿主细胞中被中药调控的基因,并通过荧光rt-PCR对上述结果进行验证。结果:益气活血中药复方能够上调CVB3病毒性心肌炎模型大鼠心肌细胞small inducible cytokine A2(MCP-1/CCL2)、ribosomal protein S4基因的表达。结论:益气活血中药复方可能通过调节small inducible cytokine A2(MCP-1/CCL2)、ribosomal protein S4基因的表达而实现治疗病毒性心肌炎的目的。
OBJECTIVE: To observe the effect of Yiqi Huoxue Recipe on the expression of small inducible cytokine A2 (MCP-1 / CCL2) and ribosomal protein S4 gene in CVB3 viral myocarditis model rats, in order to reveal the effect of Yiqi Huoxue Compound Prescription on CVB3 Myocarditis mechanism of action, and further confirmed that traditional Chinese medicine compound Yiqi Huoxue CVB3 is an effective treatment of viral myocarditis prescription. Methods: The genes regulated by Chinese herbs were cloned by modified suppression subtractive hybridization (SSH) in host cells challenged by CVB3, and the results were verified by fluorescence rt-PCR. Results: Yiqi Huoxue Compound can upregulate the expression of small inducible cytokine A2 (MCP-1 / CCL2) and ribosomal protein S4 in CVB3 myocarditis model rats. Conclusion: The TCM compound prescription of Yiqi Huoxue may achieve the goal of treating viral myocarditis by regulating the expression of small inducible cytokine A2 (MCP-1 / CCL2) and ribosomal protein S4 gene.
