目的:观察复方灵芝降糖胶囊对2型糖尿病大鼠骨骼肌GluT-4及脂肪组织胰岛素受体mRNA基因表达的影响。方法:给予实验动物STZ,破坏一部分胰岛β细胞功能,同时饲以高脂肪饲料造成外周组织对胰岛素不敏感,两者结合诱导出接近人类2型糖尿病的动物模型,并给予不同剂量治疗4周。处死后分别测定骨骼肌、脂肪组织胰岛素受体mRNA基因表达。结果:复方灵芝降糖胶囊对骨骼肌GluT-4及脂肪组织胰岛素受体mRNA基因的表达均有提升。结论:复方灵芝降糖胶囊治疗2型糖尿病可能与其提高骨骼肌GluT-4及脂肪组织胰岛素受体mRNA基因表达从而改善胰岛素的信号传导有关。
Objective: To observe the effects of compound Lingzhi Jiangtang Capsule on mRNA expression of GluT-4 and adipose tissue in skeletal muscle of type 2 diabetic rats. Methods: STZ was given to experimental animals and the function of some islet β cells was destroyed. At the same time, the peripheral tissues were not sensitive to insulin with high fat diet. The combination of them induced the animal model close to human type 2 diabetes mellitus and treated with different doses for 4 weeks. After sacrifice, the mRNA expression of insulin receptor in skeletal muscle and adipose tissue were determined respectively. Results: Lingzhi Jiangtang Capsule increased the expression of GluT-4 mRNA and insulin receptor mRNA in skeletal muscle. Conclusion: The compound Ganoderma Hypoglycemic Capsule in treatment of type 2 diabetes mellitus may be related to the increase of GluT-4 and insulin receptor mRNA expression in skeletal muscle so as to improve the signal transduction of insulin.
