目的观察荞麦花叶黄酮(flavones of buckwheat flower and leaf,FBFL)对糖尿病大鼠血糖波动的影响。方法以4-硝基苯-α-D-吡喃葡萄糖苷(PNPG)为底物,测定不同浓度FBFL对α-葡萄糖苷酶活性的影响;用高脂饲料和小剂量链脲佐菌素(STZ)建立2型糖尿病(T2DM)胰岛素抵抗(IR)大鼠模型,用不同剂量FBFL干预6周,测定大鼠随机血糖、餐后血糖的动态变化,禁食24 h内血糖波动及糖耐量情况。结果 FBFL对α-葡萄糖苷酶活性有明显的抑制作用,IC50为4.205 mg/L,与阿卡波糖(ACAR)的效价比约为15.24∶1;FBFL能降低糖尿病大鼠随机和餐后血糖值(P<0.05),并改善糖耐量,有明显的剂量和时间依赖性。结论 FBFL可降低糖尿病大鼠随机和餐后血糖值,对血糖波动有稳定作用,其机制可能是通过抑制α-葡萄糖苷酶活性实现的。
Objective To observe the effects of flavones of buckwheat flower and leaf (FBFL) on blood glucose in diabetic rats. Methods The effects of different concentrations of FBFL on α-glucosidase activity were determined by using 4-nitrobenzene-α-D-glucopyranoside (PNPG) as substrate. High fat diet and low dose streptozotocin STZ) to establish T2DM insulin resistance (IR) rat model and intervention with different doses of FBFL for 6 weeks. The changes of random blood glucose, postprandial blood glucose, fasting blood glucose and glucose tolerance within 24 h of fasting . Results FBFL had a significant inhibitory effect on the activity of α-glucosidase with an IC50 of 4.205 mg / L and a titer of about 15.24:1 with acarbose (ACAR). FBFL could reduce the effects of both random and postprandial Blood glucose (P <0.05), and improve glucose tolerance, a significant dose and time-dependent. Conclusion FBFL can reduce the random and postprandial blood glucose levels in diabetic rats, which has a stabilizing effect on the fluctuation of blood glucose. The mechanism may be through the inhibition of α-glucosidase activity.
