目的 观察 3T3 L1前体脂肪细胞诱导分化过程中Ⅱ型钙调蛋白激酶δ亚基 (CAMKⅡD)基因表达水平的变化 ,探讨CAMKⅡD基因与肥胖发生的关系。方法 体外培养 3T3 L1前体脂肪细胞 ,在诱导 3T3 L1脂肪细胞分化成熟的不同时段 ,采用RT PCR技术检测脂肪细胞中CAMKⅡD基因的mRNA表达水平。结果 诱导剂刺激后 (第 0天 ) ,3T3 L1脂肪细胞中CAMKⅡD基因的mRNA表达于第 1天显著下调 ,与第 0天比较具有显著差异 (P <0 .0 1) ;第 2天 ,该基因表达水平显著上调 ,并明显高于第 0天表达水平 (P <0 .0 1) ;第 2天至 3T3 L1细胞分化为成熟脂肪细胞 (第 10天 ) ,该基因表达一直维持在较高水平 (P >0 .0 5 )。结论 CAMKⅡD基因参与了脂肪细胞分化的调控 ,与肥胖发生相关 ,其在3T3 L1细胞分化过程中的表达变化可能有利于脂肪细胞的分化成熟和脂质积聚
Objective To observe the changes of CAMK Ⅱ D gene expression during 3T3 L1 preadipocyte differentiation and to explore the relationship between CAMK Ⅱ D gene and obesity. Methods 3T3 L1 preadipocytes were cultured in vitro, and the mRNA expression of CAMKⅡD in adipocytes was detected by RT-PCR at different time points when 3T3 L1 adipocytes were induced to differentiate. Results The mRNA expression of CAMKⅡD gene in 3T3 L1 adipocytes was significantly down-regulated on day 1 after induction with inducer (day 0), and significantly different from that on day 0 (P0.01). On day 2, The gene expression level was significantly up-regulated and significantly higher than that on the 0th day (P <0.01). From day 2 to 3T3 L1 cells differentiated into mature adipocytes (day 10), the gene expression was maintained at a higher level Level (P> 0.05). Conclusions The CAMKⅡD gene is involved in the regulation of adipocyte differentiation, which is related to the occurrence of obesity. The change of CAMKⅡD gene expression in 3T3 L1 cells may be beneficial to the differentiation and fat accumulation of adipocytes and lipid accumulation
