目的:筛选并鉴定胰腺癌组织中的差异表达蛋白,并探讨其临床意义。方法:应用双向凝胶电泳技术对12对胰腺癌组织和癌旁组织样品进行分析,筛选出差异表达蛋白质;应用MALDI-TOF-MS/MS技术对差异表达蛋白质进行鉴定。结果:优化的胰腺组织双向凝胶电泳技术具有良好的分辨率和可重复性;应用双向凝胶电泳技术筛选出胰腺癌组织中30个差异表达蛋白质;应用MALDI-TOF-MS/MS技术鉴定了胰石蛋白、钙囊素、热休克蛋白B6等24个蛋白质,其中15个蛋白质在胰腺癌组织中表达上调,9个蛋白质表达下调。结论:双向凝胶电泳、生物质谱等蛋白质组学技术在胰腺癌的基础研究中有重要价值和应用前景,有助于筛选蛋白标志物和阐明分子发病机制。胰腺癌组织中有多个异常表达蛋白质,这些蛋白质与胰腺癌的发生相关,可能成为胰腺癌蛋白标志物和药物治疗的靶蛋白。
Objective: To screen and identify differentially expressed proteins in pancreatic cancer and to explore its clinical significance. METHODS: Twelve pairs of pancreatic cancer tissues and para-cancerous tissue samples were analyzed by two-dimensional gel electrophoresis and screened for differentially expressed proteins. The differentially expressed proteins were identified by MALDI-TOF-MS / MS. RESULTS: The optimized pancreatic tissue two-dimensional gel electrophoresis technique had good resolution and repeatability. Thirty differentially expressed proteins in pancreatic cancer tissues were screened by two-dimensional gel electrophoresis. The expression of 30 differentially expressed proteins in pancreatic cancer tissues was identified by MALDI-TOF-MS / MS 24 proteins such as pancreatic protein, calcium-capping hormone and heat shock protein B6, among which 15 proteins were upregulated in pancreatic cancer and 9 proteins were down-regulated. Conclusion: Proteomics techniques such as two-dimensional gel electrophoresis and bio-mass spectrometry have important value and application prospect in the basic research of pancreatic cancer, which can help to screen the protein markers and clarify the molecular pathogenesis. There are several abnormally expressed proteins in pancreatic cancer tissues. These proteins are correlated with the occurrence of pancreatic cancer and may be the target proteins for pancreatic cancer protein markers and drug therapy.
