目的 观察人重组胰岛素样生长因子 1(rhIGF 1)对糖尿病大鼠肾脏的影响。方法 采用生化法、放射免疫法及逆转录 聚合酶链反应 (RT PCR)技术等方法。结果 (1)糖尿病治疗组 2 4h尿白蛋白排泄率和尿量低于糖尿病对照组 ;(2 )糖尿病对照组血清IGF 1含量、肾组织IGF 1含量及IGF 1信使核糖核酸 (IGF 1mR NA)表达明显低于正常对照组 ;糖尿病治疗组血清IGF 1含量明显高于糖尿病对照组 ,肾组织IGF 1含量及IGF 1mRNA表达与糖尿病对照组无明显差异 ;(3)糖尿病对照组血清生长激素 (GH)高于正常对照组 ,糖尿病治疗组血清GH低于糖尿病对照组 ;(4)电镜观察大鼠肾脏病理切片发现rhIGF 1对糖尿病大鼠肾脏损害有改善作用。结论 小剂量外源性rhIGF 1不会加重糖尿病肾脏的损害。
Objective To observe the effects of recombinant human insulin like growth factor 1 (rhIGF 1) on the kidneys of diabetic rats. Methods Biochemical methods, radioimmunoassay and reverse transcription polymerase chain reaction (RT PCR) techniques. Results (1) Urine albumin excretion rate and urinary output were lower in diabetic group than those in control group at 24 hours. (2) Serum IGF1 level, renal IGF1 level and IGF1mRNA in diabetic control group The expression of IGF1 in diabetes mellitus group was significantly higher than that in the diabetic control group. There was no significant difference in the expression of IGF1 and the expression of IGF1mRNA between the diabetes mellitus group and the diabetic control group. (3) The levels of serum growth hormone ) Was higher than that of the normal control group. The serum GH of diabetic patients was lower than that of diabetic control group. (4) Electron microscopy showed that rhIGF 1 could improve renal damage in diabetic rats. Conclusion Low-dose exogenous rhIGF-1 does not aggravate diabetic nephropathy.
