目的:建立毛细管气相色谱法测定喷昔洛韦乳膏中三氯叔丁醇含量。方法:采用气相色谱法直接进样,进样体积为1μl;色谱柱为DB-624毛细管柱(30 m×0.32 mm,1.8μm);载气为氮气;流速为1.5 ml·min~(-1)。FID检测器,检测器温度为250℃;进样口温度为200℃,柱温采取程序升温:初始温度50℃,维持5 min,以30℃·min~(-1)升温至170℃,维持4 min。以乙腈为溶剂,采用内标法测定喷昔洛韦乳膏中三氯叔丁醇含量。结果:三氯叔丁醇在40.0~200.0μg·ml~(-1)浓度范围内线性关系良好(r=0.999 8),平均回收率为99.8%(RSD=0.6%,n=9)。结论:该法简便、重复性及准确性好,可用于喷昔洛韦乳膏中三氯叔丁醇含量测定。
Objective: To establish a method for the determination of chlorobutanol in penciclovir cream by capillary gas chromatography. Methods: Direct injection was performed by gas chromatography. The injection volume was 1μl. The column was DB-624 capillary column (30 m × 0.32 mm, 1.8 μm). The carrier gas was nitrogen and the flow rate was 1.5 ml · min -1 ). FID detector with detector temperature of 250 ° C and inlet temperature of 200 ° C. The column temperature was programmed to ramp: the initial temperature was 50 ° C and maintained for 5 min. The temperature was raised to 170 ° C at 30 ° C min -1 4 min. Using acetonitrile as solvent, the content of chlorobutanol in penciclovir cream was determined by internal standard method. Results: The calibration curve was linear over the concentration range of 40.0 ~ 200.0μg · ml -1 (r = 0.999 8). The average recovery was 99.8% (RSD = 0.6%, n = 9). Conclusion: The method is simple, reproducible and accurate, and can be used for the determination of chlorobutanol in penciclovir cream.
