以紫荆花粉为试材,利用离体培养法、碘-碘化钾染色法(I2-KI)、醋酸洋红染色法和氯化三苯基四氮唑(TTC)染色法对花粉生活力进行测定,并研究了不同贮藏条件对花粉萌发的影响,以期了解紫荆花粉的萌发特性及适宜的贮藏方法。结果表明:培养基中添加不同浓度的蔗糖和硼酸对花粉的萌发有促进作用,紫荆花粉萌发的最适培养基为15%蔗糖+0.004%硼酸+0.08%的琼脂,萌发率达88.46%;I2-KI染色法和TTC染色法不适用于测定紫荆花粉生活力;而醋酸洋红染色法可以作为测定紫荆花粉生活力的方法;花粉生活力随贮藏时间的延长而下降,最佳贮藏方法为4℃干燥,耐贮藏力超过26d。
Using Bauhinia pollens as test material, pollen viability was determined by in vitro culture, iodine - potassium iodide staining (I2-KI), acetic acid magenta staining and TTC staining. The effects of different storage conditions on pollen germination were studied in order to understand the germination characteristics and suitable storage methods of Bauhinia pollen. The results showed that adding different concentrations of sucrose and boric acid to the medium promoted the germination of pollen. The optimum culture medium for Bauhinia pollen germination was 15% sucrose + 0.004% boric acid + 0.08% agar and the germination rate was 88.46%. I2 -KI staining method and TTC staining method are not suitable for the determination of Bauhinia pollen viability; and acetic acid magenta staining method can be used as a measure of Bauhinia pollen viability; pollen viability decreased with storage time, the best storage method for 4 ℃ Dry, resistant to storage more than 26d.
