目的:探讨肿瘤抑制基因PTEN及血管内皮生长因子(VEGF)及其受体FLT1(VEGFR1)在白血病中的表达及其相关性。方法:应用实时荧光定量PCR(FQ-PCR)检测50例初治急性髓系白血病(AML),10例缓解期AML,10例慢性粒细胞白血病慢性期(CML-CP)、10例CML急变期(CML-BC)患者和10例正常人骨髓单个核细胞(MMNCs)中PTEN、VEGF、FLT1 mRNA的表达。结果:初治AML患者PTEN mRNA表达水平明显低于缓解组AML和正常对照组,CML-BC患者PTEN mRNA表达水平低于CML-CP患者,在AML初治白血病中PTEN mRNA与VEGF和FLT1 mRNA表达负相关,PTEN低表达、VEGF和FLT1高表达与AML缓解率及髓外浸润相关。结论:髓系白血病患者中低表达PTEN基因,高表达VEGF及FLT1基因,并与白血病预后不良及髓外浸润密切相关,三者相互作用共同参与了白血病的发病。
Objective: To investigate the expression of tumor suppressor gene PTEN, vascular endothelial growth factor (VEGF) and its receptor FLT1 (VEGFR1) in leukemia and its correlation. Methods: Fifty cases of newly diagnosed acute myeloid leukemia (AML), 10 cases of remission AML, 10 cases of chronic myeloid leukemia (CML-CP) and 10 cases of CML were detected by real-time fluorescence quantitative PCR (CML-BC) and 10 normal human bone marrow mononuclear cells (MMNCs). Results: The expression of PTEN mRNA in newly diagnosed AML patients was significantly lower than that in AML and normal controls. The expression of PTEN mRNA in CML-BC patients was lower than that in CML-CP patients. The expression of PTEN mRNA, VEGF and FLT1 mRNA in AML naive leukemia patients Negative correlation, low expression of PTEN, high expression of VEGF and FLT1 correlated with the remission rate of AML and extramedullary infiltration. CONCLUSION: The low expression of PTEN gene in myeloid leukemia patients with high expression of VEGF and FLT1 gene is closely related to the poor prognosis of leukemia and extramedullary infiltration. The three interactions are involved in the pathogenesis of leukemia.
