目的:建立BALB/c小鼠肝癌动物模型,观察分析肿瘤转移相关蛋白1(MTA1)在建模过程中表达的变化及可能的意义。方法:采用联合二乙基亚硝胺(DEN)/四氯化碳(CCl4)/乙醇的方法诱导正常成年BALB/c雄性小鼠150 d,HE染色观察小鼠肝脏病变情况,免疫组化染色观察MTA1在肝脏病变部位的表达情况。结果:实验组小鼠肝脏镜下依次出现炎症、纤维化和肿瘤性改变。在肝脏病变进展的进程中,MTA1的表达量增加;且肝硬化期,MTA1主要在胞质中表达。结论:MTA1在DEN诱发小鼠肝肿瘤过程中表达量增加、表达位置改变,提示MTA1可能在肝脏肿瘤形成的全程均具有重要作用。
OBJECTIVE: To establish an animal model of liver cancer in BALB/c mice and observe the changes in the expression of metastasis-associated protein 1 (MTA1) in the modeling process and its possible significance. METHODS: Normal adult BALB/c male mice were induced with diethylnitrosamine (DEN)/carbon tetrachloride (CCl4)/ethanol for 150 days. Liver lesions were observed by HE staining and immunohistochemical staining was performed. Observe the expression of MTA1 in liver lesions. RESULTS: In experimental mice, liver inflammation, fibrosis, and neoplastic changes were observed. In the course of liver disease progression, the expression of MTA1 increases; and in liver cirrhosis, MTA1 is mainly expressed in the cytoplasm. Conclusion: The expression of MTA1 increased in the process of DEN-induced mouse liver tumors, and the expression of MTA1 changed. It suggested that MTA1 may play an important role in the development of liver tumors.
