目的研究大孔树脂吸附法富集和纯化白花刺参总皂苷的工艺参数及条件。方法以总皂苷为考察指标,对HPD-100、HPD-300、HPD-722、D-101、SA-2、AB-8、ADS-7、X-5 8种大孔吸附树脂富集和纯化总皂苷的吸附和解吸性能进行评价。结果 HPD-100树脂纯化总皂苷的最佳条件分别为:吸附条件为药液的质量浓度为0.25 g/mL;pH值为5.32;洗脱液体积流量为1.5 BV/h;最佳吸附容量为3.4 mL/g;解吸条件为用2 BV的蒸馏水洗脱除去杂质后,换用60%乙醇4 BV洗脱。通过HPD-100树脂纯化后,白花刺参总皂苷的纯度由21.30%上升为58.19%,平均回收率为91.58%。结论建立HPD-100大孔吸附树脂富集和纯化白花刺参总皂苷的方法简便可行、专属性强,并且可以为中草药有效成分的大规模生产提供参考。
Objective To study the process parameters and conditions for the enrichment and purification of total saponins from Apostichopus japonicus by macroporous resin adsorption. Methods Total saponins were used as indexes to enrich and purify macroporous resin HPD-100, HPD-300, HPD-722, D-101, SA-2, AB-8, ADS-7 and X-5 Adsorption and desorption properties of total saponins were evaluated. Results The optimum conditions for the total saponin purification of HPD-100 resin were as follows: the adsorption conditions were as follows: the mass concentration of the liquid was 0.25 g / mL; the pH value was 5.32; the volume flow of the eluent was 1.5 BV / h; 3.4 mL / g; desorption conditions for the removal of impurities with 2 BV of distilled water, the exchange with 60% ethanol 4 BV elution. After purified by HPD-100 resin, the purity of total saponins from S. glauca increased from 21.30% to 58.19% with the average recovery of 91.58%. Conclusion The establishment of HPD-100 macroporous resin enrichment and purification of S. argenteus saponins method is simple and feasible, specific and strong, and can provide a reference for large-scale production of active ingredients of Chinese herbal medicine.
