目的研究DXS6795基因座在山西汉族人群中的遗传多态性。方法随机抽取山西汉族人群154名无关男性和96名无关女性个体血样,DNA用酚-氯仿提取,经PCR扩增,扩增产物用8%非变性聚丙烯酰胺凝胶电泳和银染法进行检测分型,对女性基因型频率分布进行Hardy-Weinberg平衡检验,并计算法医学常用参数。结果在山西汉族人群154名无关男性和96名无关女性个体中共检出4个等位基因。等位基因频率分布在0.1040-0.3960之间。χ2检验表明女性基因型频率分布符合Hardy-Weinberg平衡。女性和男性样本的个体识别率分别为0.7406和0.7047。结论DXS6795基因座在山西汉族人群中具有较高的遗传多态性,可用于法医学个人识别和亲权鉴定,并为X染色体短串联重复序列数据库的建立提供了山西汉族人群的遗传学数据。
Objective To study the genetic polymorphism of DXS6795 locus in Shanxi Han population. Methods A total of 154 unrelated males and 96 unrelated females were randomly selected from Shanxi Han population. DNA was extracted with phenol - chloroform and amplified by PCR. The amplified products were detected by 8% non - denaturing polyacrylamide gel electrophoresis and silver staining According to the Hardy-Weinberg equilibrium test, the frequency distribution of female genotypes was calculated and the common parameters of forensic medicine were calculated. Results Four alleles were detected in 154 unrelated males and 96 unrelated females in Shanxi Han population. Allele frequency distribution between 0.1040-0.3960. Chi-square test showed that female genotype frequency distribution in line with Hardy-Weinberg equilibrium. The individual recognition rates for female and male samples were 0.7406 and 0.7047, respectively. Conclusion The DXS6795 locus has a high genetic polymorphism in Shanxi Han population. It can be used for forensic identification and paternity testing. The genetic data of Shanxi Han population is provided for the establishment of X chromosome short tandem repeat database.
