目的观察茶多酚(TP)对实验性肥胖大鼠脂肪组织PPAR-γ表达的影响。方法通过高糖高脂饮食诱导建立实验性肥胖SD大鼠模型。给予TP干预10周后测定各组大鼠血糖、血脂、胰岛素、FFA水平,计算胰岛素抵抗指数(HOMA-IR)。分别用荧光定量PCR法、Western blot、免疫组化染色法检测各组大鼠脂肪组织PPAR-γ在基因及蛋白水平的表达。结果 TP组大鼠FPG、TG、TC、LDL-C、FFA、HOMA-IR均较肥胖大鼠对照组明显下降(P<0.05),TP组脂肪组织PPAR-γ在基因及蛋白水平的表达较肥胖大鼠对照组均有增加(P<0.05)。结论 TP可调控大鼠脂肪组织PPAR-γ的表达,并显著改善肥胖大鼠糖脂代谢,有望成为临床治疗肥胖及MS的有效药物。
Objective To observe the effect of tea polyphenols (TP) on the expression of PPAR-γ in adipose tissue of experimental obesity rats. Methods The experimental obese SD rat model was induced by high glucose and high fat diet. After 10 weeks of intervention, the levels of blood glucose, lipids, insulin and FFA in each group were measured and the insulin resistance index (HOMA-IR) was calculated. The mRNA and protein expression of PPAR-γ in adipose tissue of rats in each group were detected by fluorescence quantitative PCR, Western blot and immunohistochemistry respectively. Results Compared with control group, the levels of FPG, TG, TC, LDL-C, FFA and HOMA-IR in TP group were significantly decreased (P <0.05). The expression of PPAR-γ in adipose tissue of TP group was significantly lower than that of control group Obese rats increased in control group (P <0.05). Conclusion TP can regulate the expression of PPAR-γ in adipose tissue of rats and significantly improve glucose and lipid metabolism in obese rats, which is expected to be an effective drug for the treatment of obesity and MS.
