前列腺素(PG)E在气液色谱分析时不太稳定,因而在定量测定时建议把它转为较稳定的PGB。当11-脱氧PGE_1或其乙酯与稀矿酸一起加热处理时,将脱去15-羟基而形成9-氧代-13,15-二烯酸。PGE_2也发生类似的分解。在用气液色谱分析PGF_(2α)时,作者观察到同样的分解反应。在一定色谱条件下分析各种牌号的PGF_(2α)标准样品,均可测得相对保留时间分别为1min与2min的二个色谱峰。保留时间较短的峰是二(三甲基甲硅烷基)-9,11-二羟基-13,15-二烯酸甲酯。在同样条件下PGB_2对热稳定,只给出一个峰。
Prostaglandin (PG) E is not very stable at the time of gas-liquid chromatography, so it is advisable to convert it to a more stable PGB in a quantitative assay. When 11-deoxy PGE 1 or its ethyl ester is heat-treated with dilute mineral acid, the 15-hydroxyl group will be removed to form 9-oxo-13,15-dienoic acid. A similar breakdown also occurs with PGE_2. The same decomposition reaction was observed by the authors when analyzed by gas liquid chromatography for PGF_ (2α). Under certain chromatographic conditions, the two PGF_ (2α) standard samples were analyzed, and the two chromatographic peaks with relative retention time of 1 min and 2 min respectively were measured. The shorter peak retention time was bis (trimethylsilyl) -9,11-dihydroxy-13,15-dienoate. PGB_2 is thermally stable under the same conditions, giving only one peak.
