目的 :研究人类短串联重复序列D8S1179在河南汉族人群中的遗传多态性 ,并探讨该基因座在法医学和基因诊断中的应用可能性。方法 :采集河南无血缘关系汉族个体血样 ,应用Chelex法提取DNA ,聚合酶链式反应扩增 ,非变性聚丙烯酰胺凝胶电泳分型。结果 :得到D8S1179在河南汉族群体中的基因频率 ,有 8个等位基因 ,2 8个基因型 ,杂合度为 0 82 9,个体识别率为 0 95 ,非父排除率为 0 6 6。结论 :该基因座多态性较好 ,分布符合Hardy Weinberg平衡 ,可以用于个体识别和亲权鉴定。
Objective: To study the genetic polymorphism of human short tandem repeats (D8S1179) in Henan Han population and explore the possibility of using this locus in forensic medicine and gene diagnosis. Methods: Blood samples from Han unrelated Han individuals were collected. The DNA was extracted by Chelex method and amplified by polymerase chain reaction (PCR). The genotypes were determined by non-denaturing polyacrylamide gel electrophoresis. Results: The frequency of D8S1179 in Henan Han population was 8 alleles and 28 genotypes with heterozygosity of 0 82 9, individual recognition rate of 0 95 and non-parent exclusion rate of 0.6 6. Conclusion: The polymorphism of this locus is good, its distribution is in accordance with Hardy Weinberg equilibrium, and it can be used for individual identification and paternity testing.
