目的:研究新型ATP敏感性钾通道开放剂埃他卡林(IPT)对低氧性肺动脉高压(HPH)大鼠内皮素A受体(ETAR)、内皮素B受体(ETBR)mRNA表达的影响。方法:SD大鼠平均分成正常对照组、HPH组、IPT处理组(对每天缺氧饲养的大鼠给予IPT1.5mg/kg灌胃),后两组置于常压低氧舱中每天8h,每周6天,4周后逆转录多聚酶联式反应(RT-PCR)方法检测肺组织ETAR和ETBRmRNA表达。结果:与正常对照组相比,HPH组大鼠ETARmRNA表达水平明显上升(P<0.01),ETBRmRNA表达水平明显下降(P<0.01);与HPH组相比,埃他卡林干预的缺氧大鼠ETARmRNA表达明显降低(P<0.01),ETBRmRNA表达明显增加(P<0.01)。结论:HPH模型大鼠肺组织内皮素受体mRNA表达发生异常,而埃他卡林可部分逆转这种异常,有助于抑制HPH血管收缩和平滑肌增殖。
AIM: To investigate the effects of IPT, a novel ATP-sensitive potassium channel opener, on the expression of endothelin A receptor (ETAR) and endothelin B receptor (ETBR) in hypoxic pulmonary hypertensive rats . Methods: The SD rats were equally divided into normal control group, HPH group and IPT treatment group (given IPT1.5mg / kg intragastrically to rats fed hypoxia everyday), the latter two groups were placed in normobaric hypoxia chamber for 8h everyday, The expression of ETAR and ETBRmRNA in lung tissue was detected by reverse transcription polymerase chain reaction (RT-PCR) Results: Compared with the normal control group, ETAR mRNA expression in HPH group was significantly increased (P <0.01) and ETBR mRNA expression was significantly decreased (P <0.01). Compared with HPH group, ETAR mRNA expression was significantly decreased (P <0.01), ETBRmRNA expression was significantly increased (P <0.01). CONCLUSION: The expression of endothelin receptor mRNA in lung tissue of HPH model rats is abnormal, and it can partly reverse this abnormality, which can inhibit HPH vasoconstriction and smooth muscle proliferation.
