姜黄素(curcumin)是从中药姜黄中提取的酚性化合物,广泛用于食品添加剂.近年来已发现姜黄素具有多种医疗作用以及显著的防癌和抑癌作用,并发现可诱导多种癌细胞凋亡.我们在研究姜黄素诱导人胃腺癌MGC 80-3细胞凋亡时,偶然发现姜黄索在光照下(光敏化姜黄素)也可诱导细胞凋亡.本文以MGC 80-3细胞为对象,研究光敏化姜黄素诱导细胞调亡的效应,以探讨姜黄素光敏化反应在光动力学治疗肿瘤上的应用前景.以台盼蓝拒染计数法、Giemsa染色法和Hoechst 33258染色法分别观察姜黄素+光照对MGC 80-3细胞存活及细胞形态的影响;DNA琼脂糖电泳法观察姜黄素+光照诱导癌细胞DNA片断化作用;并以DNA Hoechst33258染色荧光测定法定量检测断裂及未断裂DNA含量;流式细胞仪检测术检测经姜黄素+光照处理后的细胞周期变
