目的探讨二甲双胍对细菌脂多糖(LPS)诱导下腹腔巨噬细胞分泌IL-6的影响。方法提取C57BL/6小鼠腹腔巨噬细胞并培养,采用MTT法检测不同浓度二甲双胍(1,5,10mmol/L)对腹腔巨噬细胞存活率的影响;采用ELISA法和免疫细胞化学法分别检测对照组、LPS诱导组、LPS诱导+不同浓度药物组腹腔巨噬细胞IL-6的分泌水平。结果 MTT法检测结果显示,在不同浓度二甲双胍作用下巨噬细胞存活率与对照组比较,差异无统计学意义(P=0.95)。与对照组比较,LPS诱导下腹腔巨噬细胞分泌的IL-6水平明显升高(P<0.01);与LPS诱导组比较,LPS诱导+二甲双胍组中IL-6水平降低,且IL-6水平下降程度与二甲双胍存在剂量依赖性关系(P<0.01)。结论二甲双胍能够抑制LPS诱导下腹腔巨噬细胞IL-6的分泌。
Objective To investigate the effect of metformin on secretion of IL-6 by peritoneal macrophages induced by lipopolysaccharide (LPS). Methods C57BL / 6 mice peritoneal macrophages were isolated and cultured. The effects of metformin (1,5,10 mmol / L) on the viability of peritoneal macrophages were detected by MTT assay. The levels of peritoneal macrophages were detected by ELISA and immunocytochemistry The levels of IL-6 in peritoneal macrophages in control group, LPS-induced group and LPS-induced + different concentrations of drug group were significantly higher than those in control group. Results The results of MTT assay showed that the survival rate of macrophages in different concentration of metformin had no significant difference compared with the control group (P = 0.95). Compared with the control group, the level of IL-6 secreted by LPS-induced peritoneal macrophages was significantly increased (P <0.01). Compared with LPS-induced group, the level of IL-6 in LPS-induced + metformin group was decreased and the level of IL-6 There was a dose-dependent relationship between the decline and metformin (P <0.01). Conclusion Metformin can inhibit the secretion of IL-6 induced by LPS in peritoneal macrophages.
