目的:利用葛根的ITS,psbK-psbI,trnH-psbA序列信息对不同产地葛根进行分类,为分析不同产地的葛根药材提供依据。方法:采集东北、华北、华中和西北共8个居群24份葛根样品,提取DNA,扩增ITS序列、psbK-psbI及trnH-psbA序列、测序,通过序列比对,根据差异划分基因型或单倍型。结果:ITS1,ITS2序列各有1个单碱基突变,5.8S序列有1个单碱基突变;psbK-psbI序列有2个单碱基突变;trnH-psbA序列有1个单碱基突变,还有1段10个碱基的缺失突变。结论:利用ITS序列将葛根划分成4个基因型,根据psbK-psbI和trnH-psbA序列均将葛根划分成2个单倍型,为进一步评价提供依据。
OBJECTIVE: To classify Pueraria lobata from different regions by ITS, psbK-psbI, trnH-psbA sequence information of Pueraria lobata, and to provide basis for the analysis of Radix Puerariae from different habitats. Methods: Totally 24 Pueraria lobata samples were collected from 8 populations in Northeast China, North China, Central China and Northwest China. DNA was extracted and ITS sequences, psbK-psbI and trnH-psbA sequences were amplified, sequenced and sequenced. Haplotypes. Results: One single-base mutation was found in ITS1 and ITS2 sequences, one single-nucleotide mutation in 5.8S sequence, two single-base mutations in psbK-psbI sequence, and one single-nucleotide mutation in trnH-psbA sequence. There is also a 10-bp deletion mutation. Conclusion: According to ITS sequence, Pueraria lobata can be divided into four genotypes. According to psbK-psbI and trnH-psbA sequences, Pueraria lobata can be divided into two haplotypes, which provide the basis for further evaluation.
