目的 研究运动及高脂饮食对 2型糖尿病模型—OLETF大鼠血清瘦素 (Leptin)水平的影响 ,探讨Leptin与 2型糖尿病发病之间的关系 ,以及Leptin在运动改善机体胰岛素敏感性中所起的作用。方法 对 40只自发发病的 2型糖尿病模型—OLETF大鼠和 15只LETO大鼠进行为期 9周的运动和高脂饮食干预 ,9周后用放射免疫法测定每只大鼠血清中Leptin浓度。结果 采用基础饲料饲养的OLETF大鼠非运动组和运动组血清Leptin浓度分别为 ( 6 .5 0± 0 .75 )ng/ml和 ( 1.2 8± 0 .15 )ng/ml;采用高脂饲料饲养的OLETF大鼠非运动组和运动组血清Leptin浓度分别为 ( 6 .83± 0 .6 8)ng/ml和 ( 1.6 5± 0 .37)ng/ml ;而采用基础饲料饲养的LETO大鼠非运动组和运动组血清Leptin浓度分别为 ( 1.0 2± 0 .13)ng/ml和 ( 1.2 0± 0 .14 )ng/ml;OLETF大鼠运动组血清Leptin浓度明显低于其相对应的非运动组 (P <0 .0 5 ) ,两个OLETF非运动组大鼠的血清Leptin浓度均明显高于LETO大鼠非运动组 (P <0 .0 5 ) ,而LETO大鼠血清Leptin浓度运动组和非运动组之间没有明显的差别 ,另高脂饮食对OLETF大鼠Leptin血清浓度没有明显影响。结论 Leptin可能是具有胰岛素抵抗机制的OLETF大鼠维持机体能量代谢平衡的一种调节因子 ;运动能够改善OLETF大鼠胰岛素?
Objective To investigate the effects of exercise and high-fat diet on serum Leptin level in type 2 diabetes mellitus (OLETF) rats and to explore the relationship between leptin and type 2 diabetes mellitus and the role of Leptin in exercise to improve body insulin sensitivity Role. Methods Forty spontaneous cases of type 2 diabetes mellitus (OLETF) and 15 LETO rats were randomly divided into exercise and high fat diet for 9 weeks. After 9 weeks, the serum Leptin concentrations in each rat were determined by radioimmunoassay. Results The serum leptin levels of OLETF rats fed with basal diet were (6.5 ± 0.75) ng / ml and (1.2 ± 0.15) ng / ml, respectively. The high-fat diet The serum Leptin concentrations of OLETF rats in non-exercise group and exercise group were (6 .83 ± 0.68) ng / ml and (1.6 5 ± 0.37) ng / ml respectively; while the LETO The concentrations of Leptin in the non-exercise group and exercise group were (1.02 ± 0.13) ng / ml and (1.2 ± 0.14) ng / ml, respectively. The concentration of Leptin in OLETF group was significantly lower than that of the exercise group (P <0.05). The concentrations of Leptin in two OLETF non-exercise groups were significantly higher than those in LETO non-exercise group (P <0.05), while those in LETO rats were significantly lower than those in LETO non-exercise group There was no significant difference between the exercise group and the non-exercise group, and the other high-fat diet had no significant effect on the serum concentration of Leptin in OLETF rats. Conclusion Leptin may be a regulator of energy metabolism in OLETF rats with insulin resistance mechanism. Exercise can improve insulin secretion in OLETF rats.
