目的研究曲安奈德(TA)玻璃体内注射的不同剂量和有无赋形剂对视网膜的影响。方法纯种新西兰白兔32只,随机分成4组行TA玻璃体内注射。第1组为去赋形剂的4mgTA组,第2组为去赋形剂的25mgTA组,第3组为含赋形剂的4mgTA组,第4组为含赋形剂的25mgTA组。每只动物注药前及注药后1周、1、2个月时行视网膜电图(ERG)检查,注药2个月后处死动物,摘除眼球行光学和电子显微镜检查。结果各组兔眼注药前后各时间点ERG潜伏期无明显变化,但含赋形剂组注药后ERG振幅较注药前降低,差异有统计学意义(P<0.01);光学和电子显微镜检查显示含赋形剂组视网膜有不同程度的组织结构损害。结论TA玻璃体内注射时,≤25mg的无赋形剂TA对视网膜形态和功能无影响;赋形剂的存在可导致视网膜功能和形态改变。
Objective To investigate the effect of intravitreal injection of triamcinolone acetonide (TA) on the retina with different doses and excipients. Methods Thirty-two New Zealand white rabbits were randomly divided into 4 groups with TA vitreous injection. The first group was a 4 mg TA group for the excipient, the second group was a 25 mg TA group for the excipient, the third group was an excipient-containing 4 mg TA group, and a fourth group was an excipient-containing 25 mg TA group. Electroencephalography (ERG) was performed in each animal before injection and 1 week after injection and 1 month and 2 months after injection. The animals were sacrificed 2 months after injection and optical and electron microscopic examinations were performed. Results There was no significant change in latency of ERG before and after injection in each group, but the amplitude of ERG in vehicle group was significantly lower than that before injection (P <0.01). The optical and electron microscopy Retina with excipient groups showed varying degrees of tissue damage. Conclusion TA intravitreal injection of ≤ 25 mg of vehicle-free TA has no effect on retinal morphology and function; the presence of excipients can cause retinal function and morphological changes.
