目的:探讨健脾益智胶囊对大脑中动脉栓塞(MCAO)大鼠海马脑源性神经营养因子(BDNF)、表皮生长因子(EGF)、成纤维细胞生长因子(bFGF)表达的影响。方法:建立MCAO大鼠模型,随机分为正常组、假手术组、模型组、西药组、中药组。采用免疫组化法于术后7、14、28d观察海马BDNF、EGF、bFGF表达情况。结果:术后7d,中药组BDNF、EGF、bFGF表达显著高于模型组、西药组(P<0.05)。术后14d,中药组BDNF、EGF表达显著高于模型组、西药组(P<0.05);bFGF表达与西药组无差异。术后28d,中药组BDNF、bFGF表达显著高于模型组(P<0.05),与西药组无差异;EGF表达无差异。结论:健脾益智胶囊可促进BDNF、EGF、bFGF阳性细胞数量增加,提示健脾益智胶囊能可能通过诱导BDNF、EGF、bFGF以促进缺血后神经干细胞增殖分化。
Objective: To investigate the effect of Jianpi Yizhi Capsule on the expression of brain-derived neurotrophic factor (BDNF), epidermal growth factor (EGF) and fibroblast growth factor (bFGF) in rats with middle cerebral artery occlusion (MCAO) Methods: MCAO rat model was established and randomly divided into normal group, sham operation group, model group, western medicine group and traditional Chinese medicine group. The expression of BDNF, EGF and bFGF in the hippocampus were observed by immunohistochemistry at 7, 14 and 28 days after operation. Results: At 7 days after operation, the expression of BDNF, EGF and bFGF in Chinese medicine group was significantly higher than that in model group and western medicine group (P <0.05). At 14 days after operation, the expression of BDNF and EGF in Chinese medicine group was significantly higher than that in model group and western medicine group (P <0.05). There was no difference between bFGF expression and Western medicine group. At 28 days after operation, the expression of BDNF and bFGF in Chinese medicine group was significantly higher than that in model group (P <0.05), but no difference with Western medicine group. There was no difference in EGF expression. Conclusion: Jianpi Yizhi Capsule can increase the number of BDNF, EGF and bFGF positive cells, suggesting that Jianpi Yizhi Capsule may promote the proliferation and differentiation of neural stem cells after induction of BDNF, EGF and bFGF.
