目的:探讨一种能全面、客观、准确地评价p53的表达水平的指标。方法:用免疫组化ABC法检测45例肺癌组织的p53蛋白表达,用显微图像分析仪测定阳性细胞百分率P/A(%)、平均光密度(AOD),并将前两者的乘积作为阳性水平指数(positivelevelindex,PLI)。结果:p53表达阳性率在肺鳞癌及肺腺癌分别为50%、52.2%。肺鳞癌阳性水平指数略高于肺腺癌,但两者之间差异无显著性。结论:阳性水平指数能较全面准确地反映免疫组化反应的水平,有利于不同免疫组化反应进行比较。
Objective: To explore a comprehensive, objective and accurate index for evaluating the expression level of p53. Methods: Immunohistochemical ABC method was used to detect the expression of p53 protein in 45 lung cancer tissues. The percentage of positive cells P/A (%) and the average optical density (AOD) were measured by microscopic image analyzer. The product of the former two was taken as Positive level index (PLI). Results: The positive rate of p53 expression in lung squamous cell carcinoma and lung adenocarcinoma were 50% and 52.2%, respectively. The positive index of lung squamous cell carcinoma was slightly higher than that of lung adenocarcinoma, but there was no significant difference between the two. Conclusion: The positive level index can reflect the level of immunohistochemical reaction more comprehensively and accurately, which is beneficial to the comparison of different immunohistochemical reactions.
