5-FU是临床上应用最广泛的抗肿瘤化疗药物之一,但总体疗效仅为20%~30%,其毒副反应高,疗效不理想是妨碍其更广泛采用的主要原因。多年来,人们尝试通过改变给药方法和途径、采用生物调节剂以及改变其分子结构来提高其疗效,取得了一定的效果。近年来研究发现,通过转导外源药敏基因,可提高肿瘤细胞对化疗药物的敏感性。本研究拟以尿嘧啶磷酸核糖转移酶(UPRT)基因修饰肿瘤细胞,探讨UPRT基因修饰肿瘤细胞对5-FU的增敏效应。
5-FU is one of the most widely used anti-tumor chemotherapeutic drugs in clinical practice. However, the overall efficacy of 5-FU is only 20% -30%. Its high toxicity and unsatisfactory curative effect are the main reasons that prevent its wider adoption. Over the years, people have tried to improve their curative effect by changing the method and route of administration, using biological regulators and changing their molecular structure, and achieved some results. In recent years, the study found that by transducing foreign drug sensitivity genes, can improve the sensitivity of tumor cells to chemotherapy drugs. In this study, uracil phosphoribosyltransferase (UPRT) gene modified tumor cells to explore UPRT gene modified tumor cells on the 5-FU sensitization effect.
