目的:探讨异硫氰酸苄酯对人舌鳞癌Tca8113的作用及可能作用机制。方法将体外培养的人舌鳞癌Tca8113细胞分成对照组和不同浓度异硫氰酸苄酯处理组,每组设3个复孔。采用MTT法检测不同浓度下异硫氰酸苄酯对Tca8113细胞生存率的影响;采用PI染色和Annexin V-FITC/PI双染以及流式细胞技术检测异硫氰酸苄酯作用下Tca8113细胞周期的改变和凋亡的程度;采用Western blot方法检测细胞cyclin B1和p53蛋白的表达情况。结果异硫氰酸苄酯体外能有效抑制Tca8113细胞的增殖,这种抑制作用表现出剂量依赖性;异硫氰酸苄酯能使Tca8113细胞周期停滞在G2/M期,并能诱导细胞发生凋亡,在作用过程中,细胞cyclin B1蛋白表达降低,而p53蛋白的表达增强。结论异硫氰酸苄酯能够明显抑制人舌鳞癌Tca8113细胞的增殖并能诱导其发生凋亡,该作用机制可能与p53蛋白的表达增强有关。“,”Objective To detect the effects of phenethyl isothiocyanate (PEITC) on human tongue squamous carcinoma Tca8113 cells and to investigate the possible effects. Methods Tca8113 cells were divided into different groups treated by different doses of PEITC, MTT method was used to detect the cell viability, PI staining was used to detect the cell cycle, Annexin V-FITC/PI staining was used to detect the apoptosis, and Western Blot Analysis was used to detect the expression of cyclin B1 and p53 of cells in each group. Results PEITC could inhibit the proliferation of Tca8113 cells effectively in a dose-dependent manner, cells treated with PEITC would appear a G2/M arrest during cell cycle, and the apoptotic rates of cells were significantly high in PEITC treated groups. Western Blot analysis showed a decrease in the expression of cyclin B1 and an increase in the expression of p53 of Tca8113 cells treated with PEITC. Conclusion PEITC could obviously inhibit cell growth of Tca8113 cells as well as induce cell cycle arrest and cell apoptosis, the up-regulation of p53 might be related to the mechanism.
