目的 探讨γ射线照射鼻咽癌细胞株HNE1后 ,端粒酶活性改变情况以及与端粒酶各组分基因表达的关系。方法 HNE1在γ射线照射前和照射后不同时间 ,利用RT-PCR方法检测端粒酶亚单位人端粒酶RNA (humantelomeraseRNA ,hTR)、人端粒酶相关蛋白 (humantelomerase -associated 1,TP1)和人端粒酶逆转录酶 (humantelomerasereversetranscriptase,hTERT)基因表达情况 ,以及用TelomerasePCR -ELISA方法检测端粒酶的活性。结果 HNE1在γ射线照射后端粒酶各组分中 ,hTR和TP1mRNA改变不明显 ,而hTERTmRNA逐渐降低 ,端粒酶的活性也逐渐降低。结论 HNE1经γ射线照射后 ,hTERTmRNA表达逐渐下降 ,端粒酶活性亦逐渐降低 ,hTERTmRNA改变与端粒酶活性改变呈一致性 ,提示hTERTmRNA的表达可能在调节端粒酶活性中起关键作用。
Objective To investigate the changes of telomerase activity and its relationship with the gene expression of telomerase components after irradiation of human nasopharyngeal carcinoma cell line HNE1 with γ-rays. Methods The expression of human telomerase RNA (hTR), human telomerase-associated1 (TP1) and human telomerase-associated protein 1 (hTERT1) were detected by RT-PCR before and at different time after γ- The expression of human telomerase reverse transcriptase (hTERT) gene and telomerase activity were detected by Telomerase PCR-ELISA. Results The results showed that there was no obvious change of hTR and TP1mRNA in hNE1 after γ-ray irradiation, hTERTmRNA decreased and telomerase activity decreased gradually. Conclusion The expression of hTERTmRNA gradually decreases and the activity of telomerase decreases gradually after exposure to γ-rays. The change of hTERTmRNA is consistent with the change of telomerase activity, suggesting that the expression of hTERTmRNA may play a key role in the regulation of telomerase activity.
