【目的】探讨尼可地尔对体外培养的犬心肌细胞凋亡的影响。【方法】取成功离体培养的新生犬心肌细胞,将其随机分为对照组(A 组)、过氧化氢组(B 组)、尼可地尔+过氧化氢组(C 组)、尼可地尔+5-羟基癸酸+过氧化氢组(D 组);然后通过对一系列细胞凋亡标志物的检测,探讨尼可地尔是否具有抗细胞凋亡的作用。【结果】通过采用 TUNEL 染色标记、细胞色素 c 移位、Caspase-3活化等方法显示 B 组心肌细胞凋亡率明显高于 C 组和 A 组,且尼可地尔的这种抗细胞凋亡的作用可以被 ATP 敏感钾通道阻滞剂5-羟基癸酸所阻断。B 组 Caspase-3活性随时间延长而逐渐增强,与对照组相比差异具有统计学意义(P <0.001);与 B组相比,C 组加入尼可地尔预处理后能够明显持续抑制 Caspase-3活性,差异具有统计学意义(P <0.001)。【结论】尼可地尔具有直接抗离体培养的新生犬心肌细胞凋亡的效果,其机制与线粒体上 ATP 敏感钾通道开放密切相关。“,”Objective]To explore the effect of nicorandil on the apoptosis of cultured neonatal dog cardio-myocytes in vitro.[Methods]The cultured neonatal dog cardiomyocytes were randomly divided into four groups:control group(group A);hydrogen peroxide (H 2 O 2 )group(group B);nicorandil plus H2O2group (group C);nicorandil plus 5-hydroxydecanoate (5-HD)and H 2 O 2 group(group D).Whether nicorandil can in-hibit the apoptosis of cardiomyocytes was evaluated directly by using a number of apoptotic markers.[Results]TUNEL staining,cytochrome C translocation and Caspase-3 activation showed the apoptosis rate of group B was significantly higher than that of group C and group A,and the nicorandil anti apoptotic effect could be blocked by ATP sensitive potassium channel blocker 5- hydroxy decanoic acid.The activity of Caspase-3 in group B increased gradually with time,and the difference compared with the control group was statistically significant (P <0.001);Compared with group B ,group C was treated with nicorandil pretreatment and can significantly inhibit caspase-3 activity,and the difference was statistically significant (P < 0.001 )[Conclu-sions]Nicorandil has a direct anti effect on the apoptosis of cultured neonatal dog cardiomyocytes in vitro.The mechanism is closely related of mitochondrial ATP sensitive potassium openchannel.
