目的探讨超声介导自制载紫杉醇微泡对小鼠H22皮下移植瘤的抑制效果。方法 (1)自制载紫杉醇微泡,并检测其一般特性。(2)建立小鼠H22皮下移植瘤模型,给予紫杉醇微泡+超声处理。绘制肿瘤生长曲线,计算各组抑瘤率,肿瘤行病理组织学检查,免疫组织化学法检测微血管密度(MVD),Western Blotting检测Bax、Bcl-2蛋白的表达。结果 (1)微泡呈圆形,表面光滑,平均粒径2.07μm,载药量10.13%,包封率89.58%。(2)紫杉醇微泡+超声组肿瘤生长明显减慢,肿瘤体积和质量明显下降(P<0.05),肿瘤组织HE染色呈现不同程度变性坏死,MVD表达减少,Bax的表达上调,Bcl-2的表达下调(P<0.05)。结论超声介导载紫杉醇微泡可明显抑制小鼠H22皮下移植瘤的生长,并可降低肿瘤组织MVD的表达,促进肿瘤细胞的凋亡。
Objective To investigate the inhibitory effect of ultrasound-mediated self-made paclitaxel-loaded microbubbles on mouse H22 subcutaneously transplanted tumor. Methods (1) Paclitaxel microbubbles were self-made and their general characteristics were tested. (2) Mouse H22 subcutaneous xenograft model was established, paclitaxel microbubbles + ultrasound treatment. Tumor growth curve was drawn. The tumor inhibition rate, tumor histopathological examination, MVD and Western Blotting were used to detect the expression of Bax and Bcl-2. Results (1) The microbubbles were round and the surface was smooth. The average diameter was 2.07μm, the drug loading was 10.13% and the entrapment efficiency was 89.58%. (2) The tumor growth of paclitaxel microbubble + ultrasound group was significantly slowed down, the tumor volume and quality were significantly decreased (P <0.05), the tumor tissue showed varying degrees of necrosis, the expression of MVD decreased, the expression of Bcl-2 The expression was down-regulated (P <0.05). Conclusion Ultrasound-mediated paclitaxel-loaded microbubbles can significantly inhibit the growth of mouse subcutaneous xenografts H22, and can reduce the expression of MVD in tumor tissues and promote the apoptosis of tumor cells.
