目的研究分泌型白细胞蛋白酶抑制因子(SLPI)对脂多糖(LPS)诱导的人肺上皮细胞株A549中肺表面活性物质D(SP-D)表达的影响。方法体外培养A549细胞,分为空白对照组(A组)、LPS 10μg/ml刺激组(B组)、pcDNA3.1(+)-SLPI转染组(C组)和pcDNA3.1(+)转染组(D组)。采用RT-PCR和Western blot分别检测SP-D mRNA和蛋白表达。结果与A组相比,C组SP-D mRNA和蛋白表达增加(P<0.05),而B组和D组SP-D mRNA和蛋白表达降低(P<0.05);C组SP-D mRNA和蛋白表达均较B组增加(P<0.01)。结论 SLPI能拮抗LPS的炎性作用,刺激人肺上皮细胞中SP-D的分泌,提示SLPI对LPS诱导的肺损伤具有保护作用。
Objective To investigate the effect of secreted leukocyte protease inhibitor (SLPI) on the expression of pulmonary surfactant D (SP-D) in human lung epithelial cell line A549 induced by lipopolysaccharide (LPS). Methods A549 cells were cultured in vitro and divided into blank control group (group A), LPS 10μg / ml stimulation group (group B), pcDNA3.1 (+) - SLPI transfection group (group C) and pcDNA3.1 (+) Dye group (group D). The expression of SP-D mRNA and protein were detected by RT-PCR and Western blot respectively. Results Compared with group A, the expression of SP-D mRNA and protein in group C increased (P <0.05), while the expression of SP-D mRNA and protein in group B and group D decreased (P <0.05) Compared with B group, protein expression increased (P <0.01). Conclusion SLPI can antagonize the inflammatory effect of LPS and stimulate the secretion of SP-D in human lung epithelial cells, suggesting that SLPI has a protective effect on LPS-induced lung injury.
